چکیده

Xenon derivatisation is classically performed with hydrophilic additives (e.g. glycerol) to prevent nucleation of ice when the cryoprotectant buffer containing the crystal is flash cooled. The difficulties of obtaining a suitable cryoprotectant buffer, in which crystals are stable for a period of several minutes are, however, rarely reported. Crystals of porcine pancreatic elastase have been used to demonstrate that cryoprotection using dry paraffin oil [1] or Panjelly allows with equal or better efficacy derivatisation under a xenon atmosphere prior to shock cooling [2].

MAD phasing with krypton.

Experiments demonstrating the feasibility of Kr-edge MAD on frozen crystals as a routine method for structure determination are reported. Approximately 50% of protein crystals can be successfully derivatized by pressurization with the noble gases xenon or krypton. While Xe has produced many useful derivatives for MIR phasing over the last several years, the Xe edges (K edge = 34.6 keV, L(I) = 5...

Solution of protein crystallographic structures by high-pressure cryocooling and noble-gas phasing.

Room-pressure flash-cryocooling of protein crystals is the standard way to reduce radiation damage during data collection. Typically, it is necessary to find cryoprotection conditions by trial and error, a process that is not always successful. Recently, a new method, high-pressure cryocooling, was developed that does not require penetrative cryoprotectants and typically yields very high qualit...

High-pressure cryocooling for capillary sample cryoprotection and diffraction phasing at long wavelengths.

Crystal cryocooling is usually employed to reduce radiation damage during X-ray crystallography. Recently, a high-pressure cryocooling method has been developed which results in excellent diffraction-quality crystals without the use of penetrative cryoprotectants. Three new developments of the method are presented here: (i) Xe-He high-pressure cryocooling for Xe SAD phasing, (ii) native sulfur ...

Xenon and krypton separation in a chromatographic column packed with granulated nano NaY zeolite

In order to investigate the gas separation ability of a column packed with nanozeolitic material, nano NaY zeolite was synthesized and granulated. These uniform granules packed in a chromatographic column were utilized for separation of Xe and Kr under various operating conditions. With regards to the response peaks obtained from trace injections of Xe and Kr into the column, the first and seco...

Phasing the AP2 core complex with Xe, Hg and Se.

The 200 kDa core of the heterotetrameric AP2 clathrin adaptor complex was phased with xenon, mercury and selenomethionine derivatives. The phasing has been analysed in retrospect to determine how many of the derivative data sets were necessary for optimum phasing and what features were most useful in deciding which derivatives would make a positive contribution. The relative contributions of th...
